Highly sensitive colorimetric assay compatible with detergent solubilized protein solutions  

The BCA method is faster and easier than Lowry method, ease of use with much greater tolerance to interference from nonionic detergents and buffer salts. In particular, it is insensitive to detergents such as Triton-100 and SDS (1% ). The principle of the bicinchoninic acid (BCA) assay is the formation of a Cu2+-protein complex under alkaline conditions, followed by reduction of the Cu2+ to Cu1+, in a temperature dependent reaction. The amount of Cu2+ reduced is proportional to the amount of protein present in the solution. Next, two molecules of bicinchoninic acid (BCA) chelate with each Cu+ ion, forming a purple-colored product that strongly absorbs light at a wavelength of 562 nm that is linear for increasing protein. The amount of protein present in a solution can be quantified by measuring the absorption spectra and comparing with protein solutions with known concentrations.

RUBY Gel Stain is a ready to use kit for rapid and sensitive protein staining in 1 [caption id="attachment_5250" align="alignright" width="231"] Ruby Fluorescent stain for protein gel stainin[/caption] D and 2D SDS polyacrylamide electrophoresis gels. It enables optimal visualization and quantitation of proteins with high contrast and the same sensitivity as silver staining without its drawbacks. The staining procedure is a simple 220 minutes, three step protocol. After staining, proteins can be removed from the gel and analyzed by mass spectrometry without interference from the stain. The dye has optimal excitation at 302 and 470 nm, with an emission maximum at approximately 610 nm.
